Sampling sites

Polluted site: From a pine forest soil in Harjavalta (southwestern Finland). The area is mainly polluted by Cu from a Cu-Ni smelter. Average Cu content at the site was 2,000 mg/kg of dry soil, and bioavailable Cu content was 150 mg/kg of dry soil.
Unpolluted site: Forest stand near the city of Lahti (southern Finland).

Test system

Cu resistance in the asexual worms originating from two site (uncontaminated and contaminated)

Strains & soil samples

Collected from a pine forest in Harjavalta, southwest Finland, in October 1996. The area is mainly polluted by Cu from a Cu-Ni smelter. Metal-stressed worms were collected 2 km (Polluted sites) and reference worms (Unpolluted sites) 8 km from the smelter.

Test design

Resistance assay (the filter paper contact test (OECD, 1984)).
Individuals of similar size, body length 7-8.3 mm, width 0.15-0.3 mm and number of segments 39-44.

Polluted sites: CuSO4 (0, 1, 1.5, 2.25, 2.8, 3.375 and 5.1 mg Cu m-2; n=5) 20+/-2 degrees C in darkness for 96 hr.
Unpolluted sites: CuSO4 (0, 500, 1000, 1500, 2000, 2500 and 3000 mg kg-1 of dry soil; n=5) 20+/-2 degrees C in darkness for 2 weeks.

Measurements/observations

Body length and width (10th segment from the head) of adult individuals (>35 segments), number of segments, density of worms.

Evaluations

LC50-values by trimmed Spearman-Karber method.
