Glucose phosphate isomerase (GPI) was extracted from cultures of the L1 strain of Naegleria gruberi and examined using specific enzyme staining following electrophoresis in starch gel. Two isoenzymes were found, whose relative proportions varied from preparation to preparation. "Smooth" (S) and "Rough" (R) cysts were isolated from the culture. The clones retained S or R characteristics through more than ten passages, although behaving otherwise as N. gruberi isolates. A "fast" GPI isoenzyme was extracted from the S clone, and a "slow" isoenzyme from the R clone. GPI from an S clone of N. gruberi from the Cambridge collection (1518/1e) was of apparently identical electrophoretic mobility to that of the S clone of L1. The taxonomic significance of these findings is discussed.
