Freezing and thawing

Test design
- Freezing: Controlled freezing was carried out in a commercial Linde BF-3 unit. The rotifers were collected in small droplets (about 0.05 cc) which were placed on 6-mm diameter (0.25 mm thick) copper disks. Cooling rate (degrees C/min) 1, 2, 3, 4, 5, 6, 7, 8, 9, 10 and very rapid cooling. Pretreatment with Glycerol (%) No pretreatment, 0.1, 2.5, 5, and 10. Storage period (days) 0, 2, 10 and 20.
- Thawing: All specimens were quickly thawed by plunging the copper disks into depression dishes containing 2 to 3 ml of nutrient medium at room temperature.
Measurements/observations
Recovery from freezing was scored in these depression dishes 18 to 24 hrs after thawing.

A single rotifer on a slide glass was subjected to desiccation. After each desiccation time, water added the slide glass.
Temperature 25-27 degrees, relatively humindity 68-70%.
