The role of microtubules was investigated in the conjugation process of the hypotrich ciliate Oxytricha bifaria by using colchicine and vincristine. The lowest drug concentrations used (COL: 1.8 mg/ml, VCR: 1.2x10-2 mg/ml), which proved to be uneffective on the growth rate, partially prevented cell conjugation, while higher dosages (COL: 10 mg/ml, VCR: 0.1 mg/ml) not only totally inhibited cell conjugation but also split early couples. The effects of the two inhibitors were found to be completely reversible. The possible mechanism of action of the drugs is considered.
