Colchicine (COL)/0, 1.8, 3.6, 5.5, and 10 mg/ml. Vincristine (VCR)/0, 1.2x10E-2, 5x10E-2, and 10E-1 mg/ml.
Test design
Temperature 21+/-1 degrees C.
- Cell cycle: Dividers from exponentially growing culture were individually isolated in depression slides to which (1) freshly prepared food was added, (2) fresh food + COL or VCR.
- Conjugation: Cells were starved for 24 h and mixed in a physiological solution (SMB) (control cells), in COL or VCR dissolved in SMB.

Test design
O. granulifera was grown in Pringsheim's medium with Chlorogonium elongatum as food and treated, after 2 days starvation, with 50 uM Cd (5.58 ug Cd/ml). Cells were harvested by centrifugation at 2,000 g on the first day after Cd administration and homogenized in the cold. The homogenate was centrifugated at 100,000 g for 1 hr 30 min at 5 degrees C. Cd-chelating proteins were purified by a combination of gel filtration and ion-exchange chromatography.
