Test design

The test is performed routinely in 30 ml crystal polystyrene screw-capped vials. If products tested can adsorb on plastic or alter it, borosilicate glass or teflon vials can be used. The vials are prepared for the test as follows:
- Toxicant solution in MM (MM medium composition: CaCl2/2H2O 107 mg, NaCl 14.5 mg, NaNo3 4.5 mg, MgSO4/7H2O 75.7 mg, Na2SO4 39.5 mg, NaHCO3 135 mg, distilled, de-ionized or Milli-Q grade water added to 1000 ml, pH 8.15+0.02) 4 ml. 1.25 final concentration in the vial.
- Escherichia coli 0.25 ml. Strain ATCC 11303 (Sigma EC11303), strain ATCC 9637 (Sigma EC9637) or strain K12 (Sigma EC1), suspension 2.5 mg/ml in MM medium.
- C. campylum dilution, 3333 cells/ml 0.75 ml. The final volume is 5 ml and initial cell concentration is 500 cells/ml.
The vials are incubated in the dark at 28 degrees C for 24 hr. At the end of the incubation, each flask is fixed with 1 ml of 5% glutaraldehyde solution.

Strains

The organisms was obtained, initially, from Stirone Stream (northern Italy), a freshwater environment free from known toxicants and, in particular, from heavy metals. The culture medium was constituted of boiled rice and wheat grain in 10 ml of filtered natural water. 20+/-1 degrees C, photoperiod of 16:8 hr light:dark.

Toxicant/concentrations

NiCl2/6H2O (1.12, 1.25, 1.4, 1.5 ppm).

Test design

Tissue culture plates with 24 wells, 3 replicates.

Measurements

Mortality or the survivorship.
