Improvements in fixation and cell handling, chemical dehydration with DMP (2,2-Dimethoxypropane) and the first-time use of Dimethyl-sulfoxide as a dehydratant, make it possible to obtain large quantities of critical-point dried Amoeba proteus showing neither surface deformations nor microvilli. A new dry-cleaving technique allows SEM examination of internal parts of the cell, in particular the contractile vacuole and its associated organelles at various stages of its cycle. The precise mode of vacuolar membrane vesiculation at systole is described.
